Mechanism of Action
Oxytocin is a cyclic nonapeptide hormone and neuropeptide that primarily acts through binding to the oxytocin receptor (OXTR), a class A G protein-coupled receptor coupled to Gq proteins, leading to activation of phospholipase C-β, hydrolysis of PIP2, generation of IP3 and DAG, intracellular Ca²⁺ mobilization, and downstream modulation of protein kinase C and other signaling cascades. This facilitates processes such as smooth muscle contraction, enhanced social cognition via central pathways, anti-inflammatory effects through NF-κB inhibition and reduced pro-inflammatory cytokine release (e.g., TNF-α, IL-6), mitochondrial protection against oxidative stress, and promotion of tissue repair and resilience. Recent laboratory investigations highlight its pleiotropic roles in modulating stress responses, supporting anti-inflammatory and antioxidant mechanisms, influencing metabolic homeostasis, and exerting neuroprotective effects in models of adversity, aging, and dysfunction, offering reassuring evidence of its broad adaptive influence on physiological balance and recovery.
